This study sheet presents specific analytical methods we developed for the characterisation of PEGylated proteins. It covers molecular mass distribution and PEG‑to‑protein ratio determination by MALDI‑TOF mass spectrometry and SEC‑UV/RI/MALS, conjugation site determination by peptide mapping using UPLC‑UV/MS, and free PEG quantification using HPLC with charged aerosol detection.
